General description

Paired amphipathic helix protein Sin3a (UniProt Q60520; also known as Histone deacetylase complex subunit Sin3a, mSin3a, Transcriptional co-repressor Sin3a) is encoded by the Sin3A (also known as mSin3A, AW553200, Kiaa4126, mKIAA4126) gene (Gene ID 8819) in murine species. The transcription regulator Sin3a is a scaffold protein that interacts with various proteins to form specialized complexes that mediate different biological processes, including chromatin modification, embryonic development, DNA repair, mitochondrial metabolism, and gene transcription. SIN3a harbors four paired α-helices (PAH), a histone interaction domain, and a highly conserved region. The histone interaction domain mediates HDACs binding, whereas the highly conserved region interacts with proteins necessary for repression, such as the nuclear receptor hormone co-repressor Alien. The four PAHs represent the main interacting domains of SIN3a and mediate the interactions with a wide variety of DNA-binding transcription factors. Members of the Mad family of repressor proteins, for example, specifically interact with the PAH2 through their Sin3 interacting domain (SID). Although widely studied for its involvement in transcriptional repression, Sin3a is shown to be essential for aryl hydrocarbon receptor-mediated induction of gene transcription.
