Analysis Note

ControlIncluded Positive Antigen: Catalog # 12-309, HeLa nuclear extract. Add an equal volume of Laemmli reducing sample buffer to 5-10 µL of lysate and boil for 5 minutes to reduce the preparation. Use 20 µg per lane for minigels.

Application

Use Anti-p21/WAF1/Cip1 Antibody (Mouse Monoclonal Antibody) validated in WB, IP to detect p21/WAF1/Cip1 also known as CDK-interacting protein or cyclin-dependent kinase inhibitor 1A.

Research CategoryEpigenetics & Nuclear Function

Immunoprecipitation: 4 µg of a previous lot immunoprecipitated Cip1 from 500 µg of HeLa nuclear extract.

Research Sub CategoryCell Cycle, DNA Replication & Repair

General description

Two families of cyclin dependent kinase inhibitors (CKIs) have been identified. The p21WAF1/Cip1 family inhibits all kinases involved in the G1/S transition. The p16INK4a family inhibits Cdk4 and Cdk6 specifically. The biochemical activities and patterns of expression of CKIs during development implicate these proteins as the primary effectors of signaling pathways that control cell cycle exit, an event that is critical for differentiation. Studies have shown that p21WAF1/Cip1 binds tightly to the G1 and S phase kinases, cyclin E/Cdk2, cyclin D/Cdk4, and cyclin A/Cdk2, and effectively inhibits their activity, whereas p21WAF1/Cip1 is a relatively poor inhibitor of the G2/M phase kinase cyclin B/Cdc2. In addition, p21WAF1/Cip1 inhibits proliferating cell nuclear antigen (PCNA), binds to and inhibits inactivation of Rb, which is essential for cell cycle progression, and may also interact with E2F and protect cells against p53-mediated apoptosis.
